چکیده

18 Bacterially-mediated fluxes of energy and matter are dynamic in time and space coupled with 19 shifts in bacterial community structure. Yet, our understanding of mechanisms shaping 20 bacterial biogeography remains limited. Near-surface seawater was collected during transits 21 between Honolulu and Station ALOHA in the North Pacific Subtropical Gyre to examine the 22 shape of occupancy-frequency distributions (the different number of populations occupying 23 different number of sites) and determine bacterial metapopulation dynamics. Bacterial 16S 24 rRNA gene amplicons were sequenced from whole seawater and filter-size fractionated 25 plankton DNA samples while also separating the community into distinct taxonomic groups 26 at phyla/class and analyzing these compartments separately. For the total seawater (i.e. the 27 >0.2 μm size fraction) and picoplankton communities (i.e. the size fraction >0.2 μm and < 3.0 28 μm), but not the large size fraction community (i.e. the >3.0 μm size fraction), most 29 individual operational taxonomic units (OTUs) occupied a single site and the number of 30 OTUs occupying different number of sites followed a significant bimodal pattern with several 31 core OTUs occupying all sites. Nevertheless, only Cyanobacteria (in particular 32 Prochlorococcus sp.) and in a few instances also Alphaproteobacteria (in particular SAR11 33 . Quantifying subtropical North Pacific gyre mixed layer primary productivity from Seaglider observations of diel oxygen cycles

Using autonomous underwater gliders, we quantified diurnal periodicity in dissolved oxygen, chlorophyll, and temperature in the subtropical North Pacific near the Hawaii Ocean Time-series (HOT) Station ALOHA during summer 2012. Oxygen optodes provided sufficient stability and precision to quantify diel cycles of average amplitude of 0.6μmolkg . A theoretical diel curve was fit to daily observat...

Light dependence of [3H]leucine incorporation in the oligotrophic North Pacific ocean.

The influence of irradiance on bacterial incorporation of [(3)H]leucine was evaluated at Station ALOHA in the oligotrophic North Pacific subtropical gyre. Six experiments were conducted on three cruises to Station ALOHA to examine how [(3)H]leucine incorporation varied as a function of irradiance. Two experiments were also conducted to assess the photoautotrophic response to irradiance (based o...

Stable nitrogen isotope dynamics of dissolved nitrate in a transect from the North Pacific Subtropical Gyre to the Eastern Tropical North Pacific

The stable nitrogen isotopic composition of nitrate, concentrations of inorganic nitrogen and phosphorus, dissolved oxygen and nitrification rates were determined at six stations ranging from the oligotrophic North Pacific Subtropical Gyre (NPSG) to the more productive Eastern Tropical North Pacific (ETNP). Nitrification rates increased along the transect from a maximum rate of 1 nmol L 1 d 1 a...

Experiments linking nitrogenase gene expression to nitrogen fixation in the North Pacific subtropical gyre

We examined diazotroph activity in controlled experiments containing natural populations of nitrogen (N2)fixing microbes collected offshore from Kane’ohe Bay, Hawai’i, and from Station ALOHA (A Long-term Oligotrophic Habitat Assessment) in the North Pacific subtropical gyre. Quantitative polymerase chain reaction and reverse transcription-polymerase chain reaction approaches showed that the abu...
